Cellular Metabolism Platform - Seahorse XFe96

Operation mode and possible applications of the Seahorse XFe96 Analyzer

Quantitative analysis of the main energy producing metabolic pathway of cell populations is central for many current research questions. The two main pathways are oxidative phosphorylation (OXPHOS) in the mitochondria and anaerobic glycolysis. The Seahorse XFe96 Analyzer by Agilent can measure two parameters in a 96-well format at once: the Oxygen Consumption Rate (OCR) and the Extracellular Acidification Rate (ECAR).

OCR is a measure for the mitochondrial respiration via OXPHOS while ECAR is a measure for anaerobic glycolysis. With the respective inhibitors and /or substrates (glucose, fatty acids, keton bodies) one can furthermore determine the preferred substrate use and substance toxicity in a quantitative way. The measurements can be done in a 96-well format allowing the analysis of multiple cell lines and the effects of different chemical substances in parallel. The latter is especially important for medium- or high-throughput screening for the efficacy or toxicity of potential chemotherapeutic agents.

The devices can be used for the analysis of various cell types, such as primary cells, stem cells, cancer cell lines and immune cells. The Seahorse Analyzer can therefore help to answer manifold research questions and will help to understand the role of metabolism better.

Concept of use

The device is installed in the Institute of Biochemistry and Molecular Biology I (22.03 Floor 04). After a proper introduction by Prof. Dr. Andreas Reichert or his representatives, the device will be available to use for all employees of the HHU, UKD and BMFZ. For this, a web based calendar is installed to allow booking of a time slot online. Access will be granted after introduction in our institute. All material has to be provided by the users.
