Liquid Biopsy

In recent years, medicine has focused more and more on the use of cancer material circulating in the blood - the so-called liquid biopsy. Among them are the circulating cell-free DNA (cfDNA) and exosomes secreted by apoptotic and necrotic tumor cells, as well as tumor cells (circulating tumor cells, CTCs) themselves, which are derived from the primary tumor and/or metastases. The blood is taken in a minimally invasive manner, does not require any surgical intervention and can therefore be implemented cost-effectively at shorter intervals. Likewise, the potential of the "liquid biopsy" consists in checking the response to therapy and detecting a possible recurrence at an early stage and thus achieving more effective treatment of the patient.

Projects:

  • New methods to detect and isolate rare cells from body fluids
  • Single cell transcriptome analysis of CTCs during therapy
  • Analysis of endocrine resistance in breast cancer CTCs on DNA and protein level
  • Detection and analysis of circulating endometrial cells
  • Functional analysis of CTCs in a Zebrafish transplantation model

Methods:

  • Antibody-based CTC detection and enrichment (CellSearch)
  • Microfluidic isolation of CTCs (Parsortix)
  • Automated single cell micromanipulation (CellCelector)
  • Multiple fluorescence microscopy
  • Single cell DNA and RNA analysis
